General description

SNX2 (sorting nexin 2) is a member of cytoplasmic and membrane proteins called SNXs. This protein is expressed in thyroid in the columnar and active follicular cells. It contains the PX (phox homology) domain, found in all SNX members, which binds to phosphatidylinositol 3-phosphate (PI3P). It also contains a BAR domain adjacent to its PX domain, towards the C-terminal. It is thought to form a dimer with other SNX member proteins, which in turn make a part of the heteropentameric complex called retromer. SNX2 also contains a coiled-coil domain in its C-terminal. It is a homologue of vacuolar protein-sorting (Vps)5p found in yeast. It has a molecular weight of ~68kDa.

Application

Anti-SNX2 antibody produced in chicken is suitable for western blotting analysis at a dilution of 1:500, for tissue or cell staining at a dilution of 1:200.

Biochem/physiol Actions

Sorting nexin-2 (SNX2) acts as a novel marker for active thyrocytes and reflects the endosomal trafficking activity in these cells. It forms a component of the mammalian retromer complex that functions in the retrieval of the cation-independent mannose 6-phosphate receptor from endosomes to the trans-Golgi network. It is involved, though not necessary, in the lysosomal sorting of EGFR (epidermal growth factor receptor), which is internalized. Mouse embryos with inactivated SNX2 gene die mid-gestation. This study suggests that SNX2 is essential during developmental processes.
