Gene Information

Biochem/physiol Actions

Analog of pyrophosphate ion that inhibits the osteoclastic activity leading to bone resorption and osteoporosis. The compound is used in cancer research, especially in skeletal metastases and breast carcinoma. When entrapped in liposomes, it is used for macrophage-selective depletion (macrophage "suicide" technique), especially in spleen and liver. Found also to inhibit collagenase and matrix metalloproteinase1.

Interstitial collagenase present in human jaw cyst extract and purified human fibroblast-type collagenase (MMP-1) were both efficiently inhibited in vitro by clodronate, an osteoactive, antiresorptive bisphosphonate. The IC50 of clodronate to inhibit MMP-1 is 150 microM. These findings suggest an

Although macrophages are thought to be crucial for the pathogenesis of chronic inflammatory diseases, how they are involved in disease progression from simple steatosis to non-alcoholic steatohepatitis (NASH) is poorly understood. Here we report the unique histological structure termed "hepatic

Macrophages form an important cell population involved in numerous immunological processes. Techniques which eliminate these cells, in order to permit 'in vivo' studies of their function are generally imperfect and for that reason we have developed a method using the

Alcohol consumption is a major cause of liver disease. It also associates with increased cardiovascular risk and Type 2 diabetes. ALD (alcoholic liver disease) and NAFLD (non-alcoholic fatty liver disease) share pathological features, pathogenic mechanisms and pattern of disease progression.

Hematopoietic stem cells (HSCs) reside in specialized bone marrow (BM) niches regulated by the sympathetic nervous system (SNS). Here, we have examined whether mononuclear phagocytes modulate the HSC niche. We defined three populations of BM mononuclear phagocytes that include Gr-1(hi)
