General description

BCL2/adenovirus E1B 19 kDa protein-interacting protein 3 (BNIP3) is a member of the NIP3 family, a singular group of death-inducing, mitochondrial proteins. BNIP3 is comprised of several domains of interest; a highly conserved domain (CD), a C-terminal transmembrane (TM) domain, and a BH3 domain. Strongly upregulated by hypoxia, BNIP3 induces a caspase-independent cell death characterized by mitochondrial damage. Overexpression has been observed in the brain after stroke and in the adult heart following myocardial infarction. Recent studies have shown BNIP3 to be involved in cell growth inhibition through suppression of the mTOR pathway and that, when localized to the nucleus, it has the paradoxical capability of repressing apoptosis in tumors by binding the AIF expression gene.

Other homologies: Rat, equine, and ox (90% sequence homology).

Immunogen

Epitope: Internal (C-terminal half).
KLH-conjugated linear peptide corresponding to an internal sequence within the C-terminal half of human BNIP3.

Application

Anti-BNIP3 Antibody detects level of BNIP3 & has been validated for use in Immunocytochemistry and Western Blotting.
Immunocytochemistry Analysis: A representative lot detected hypoxia-induced BNIP3 upregulation in human glioblastoma-astrocytoma, epithelial-like U87 cells (Courtesy of Spencer Gibson, Manitoba Institute of Cell Biology).

Quality

Evaluated by Western Blotting in PC3 cell lysates.

Western Blot Analysis: 0.5 µg/mL of this antibody detected BNIP3 in 10 µg of PC3 cell lysate.

Target description

~60 kDa (dimer) observed. BNIP3 may be observed as a 30 kDa monomer and/or a 60 kDa dimer.

Purified rabbit polyclonal antibody in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.
