General description

CREB (cAMP response element binding protein) is a transcription factor that responds to a diverse array of stimuli including hormones, growth factors and neuronal activity. Transcriptional activation of its target genes are important for diverse cellular functions such as cell survival, growth and differentiation. Although CREB is expressed in various tissues, it plays an important regulatory role in nervous system by promoting neuronal survival, proliferation, differentiation and neurite outgrowth. In addition, CREB signaling is involved in synaptic placidity and long-term memory, and has been implicated in neurodegenerative diseases such as Alzheimer′s and Huntington′s disease.CREB is phosphorylated on Ser133 by a variety of kinases, such as PKA, CaMK, MSK, and p90RSK. Understanding the regulation and signaling by CREB in cell survival, growth and differentiation will lead to new strategies to treat cancer, neurodegenerative and other diseases. The MILLIPLEX®2-Plex Phospho/Total CREB kit contains all the components necessary to measure total CREB and phosphorylated CREB (Ser133). The MILLIPLEX® 2-Plex Phospho/Total CREB magnetic bead kit has been developed for the simultaneous detection of phosphorylated CREB (Ser133) and total CREB in a single well using the Luminex system. This detection assay is a rapid, convenient alternative to Western Blotting and immunoprecipitation procedures for the analysis of cell lysate samples.

Specificity

Cross-reactivity between the antibodies and any of the other analytes in this panel is non-detectable or negligible.

Application

Intracellular Bead-Based Multiplex Assays using the Luminex technology enables the simultaneous relative quantitation of multiple phosphorylation and total pathway proteins in tissue and cell lysate samples. Compare Multiplexing results to those of Western blotting.An overnight (4°C) incubation is recommended for best results.This assay requires 25 μL diluted, filtered cell lysate per well.This kit must be run using the provided Assay Buffer.The suggested working range of protein concentration for the assay is 1 to 25 μg of total protein/well (25 μL/well at 40 to 1,000 μg/mL).Analytes available:CREB (Total)CREB (Ser133)
