General description

The KEAP1 (kelch-like ECH-associated protein 1) gene encodes a protein containing an N-terminal BTB domain, a conserved linker domain, and a Kelch domain at the C-terminal. The gene is mapped to human chromosome 19p13.

Application

Biochem/physiol Actions

The protein Keap1 (kelch-like ECH-associated protein 1) encoded by the gene KEAP1 negatively regulates Nrf2, a bZIP transcription factor involved in the induction of phase II detoxifying and oxidative stress enzyme genes, thus protecting cells from oxidative damage and maintaining cellular redox homeostasis. The encoded protein ubiquitinates Nrf2 by forming a functional E3 ubiquitin ligase complex with Cul3 and Rbx1. It increases the rate of proteasomal degradation of Nrf2 and also alters its subcellular distribution. Loss of keap1 function leads to increase in Nrf2 levels, which results in development of resistance to chemotherapeutic drugs in lung tumors. Mutations in this gene have been observed in NSCLC (Non-small-cell lung cancer).
