General description

Cytoplasmic Isozyme

Application

Malic dehydrogenase has been used in a study to assess the use of a long-wavelength homogeneous enzyme immunoassay for the determination of amikacin in water samples. It has also been used in a study to investigate the lipid metabolism in a liver exposed to cadmium.

Biochem/physiol Actions

The surfactant cetyl (hexadecyl) trimethylammonium bromide inhibits the cytoplasmic isoform of porcine heart malate dehydrogenase but has no effect on the mitochondrial form. SDS however; inhibits both isoforms.

Unit Definition

One unit will convert 1.0 μmole of oxalacetate and β-NADH to L-malate and β-NAD per min at pH 7.5 at 25 °C.

Numerous inhibitors of cyclin-dependent kinases and glycogen synthase kinase-3 (GSK-3) are being developed in view of their potential applications against cancers and neurodegenerative disorders. Among these, paullones constitute a family of potent and apparently selective cyclin-dependent kinase and GSK-3 inhibitors.

The cationic surfactant, cetyl (hexadecyl) trimethylammonium bromide (CTAB), completely inactivates porcine heart cytoplasmic malate dehydrogenase (L-malate:NAD+ oxidoreductase, EC 1.1.1.37) at concentrations (of surfactant) which do not affect the activity of the mitochondrial isoenzyme. These concentrations are close to, or higher
