General description

Monoclonal Anti-Myosin (Light Chains 20 kDa) (mouse IgM isotype) is derived from the hybridoma produced by the fusion of mouse myeloma cells and splenocytes from an immunized mouse. Myosin is a 500,000 Da protein known to interact with identical heavy chains (200,000 daltons each) and four light chains (15,000-20,000 daltons). Myosin molecules consist of two major regions: tail (rod) and head regions.

Specificity

Monoclonal Anti-Myosin reacts best with the 20,000 dalton light chains of chicken gizzard, intestine or pig stomach myosin (SDS heat denatured and reduced).

Application

Monoclonal Anti-Myosin (Light Chains 20 kDa) antibody produced in mouse has been used in:

Monoclonal anti-myosin (Light Chains 20 kDa) antibody can be used in immunoblotting and western blotting.

Biochem/physiol Actions

Myosin is a cytoskeletal protein responsible for muscle contraction, muscle filament sliding and regulation of ATPase activity. Monoclonal anti-myosin (Light Chains 20 kDa) antibody diluted 1:200 can be used to detect MLC20. It is also useful in staining (1:10,000) 2D slab gels. Anti-myosin(Light Chains 20 kDa) antibody reacts specifically with 20 kDa light chain of chicken gizzard, chicken intestine and pig stomach myosin which is SDS, heat denatured and reduced but not with intact myosin molecule in its native state.
The two major cytoskeletal proteins implicated in cell motility are actin and myosin. Numerous investigators have shown that actin and myosin are constituents of many cell types and are involved in a myriad of cellular processes including locomotion, secretion, cytoplasmic streaming, phagocytosis and cytokinesis. Myosin molecules spontaneously assemble into filaments in solutions of physiologic ionic strength and pH. Thick filaments consist mainly of myosin molecules. Myosin is an enzyme (ATPase) and its activity is the immediate source of the free energy that drives muscle contraction. Myosin binds to the polymerized form of actin, the major constituent of the thin filament.

Physical form

The product is provided as ascites fluid with 15 mM sodium azide as a preservative.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, the solution may be frozen in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
