Specificity

Clone 3I23 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Cereblon. It targets an epitope within 19 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal region of human Cereblon.

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CRBN in Jurkat cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Cereblon (CRBN) in NIH3T3, PC3, and C6 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Cereblon (CRBN) in human cerebral cortex tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Cereblon (CRBN) in PC3 and Jurkat cells.

Anti-CRBN, clone 3I23 ZooMAb®, Cat. No. ZRB1304, is a recombinant Rabbi+J3:J17t monoclonal antibody that specifically targets Cereblon (CRBN) and is tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Protein cereblon (UniProt: Q96SW2) is encoded by the CRBN (also known s AD-006) gene (Gene ID: 51185) in human. Cereblon is peripheral membrane protein that is widely expressed with highest levels found in the brain. It is reported to play a protective role against DNA damage-induced apoptosis in cell lines and primary cells. It interacts with DNA-binding protein 1 (DDB1), Cullin 4 (Cul4A or Cul4B), and regulator of Cullins 1 (RoC1) to form the functional E3 ubiquitin ligase complex where cereblon functions as a substrate receptor of E3 ubiquitin ligase complex, which mediates the ubiquitination and subsequent proteasomal degradation of target proteins, such as MEIS2. The cullin-4 protein provides a scaffold for assembling RING box-domain protein (RBX1) and the adaptor DDB1 to form a CRL4 E3 complex, where RBX1 serves as the docking site for the activated E2 protein. The DDB1 protein recruits substrate-specificity receptors or DDB1-cullin 4-associated factors (DCAFs) to form the substrate-presenting side of the Cullin-RING E3 ubiquitin ligases complex. Cereblon has been recognized as the primary teratogenic target of thalidomide and its analogs. These drugs exert their antiproliferative effects on multiple myeloma cells and immunomodulatory effects on T cells by directly binding cereblon and promoting recruitment of the hematopoietic zinc-finger transcription factors Ikaros (IKZF1) and Aiolos (IKZF3), leading to their ubiquitination and proteasomal degradation. Mutations in CRBN gene have been linked to mental retardation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
