Specificity

Clone 1C23 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Jagged 1. It targets an epitope within 24 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal, cytoplasmic domain of human Jagged-1.

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in human kidney tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected Jagged1 in human kidney tissue sections.

Tested applications

Flow Cytometry Analysis: 0.1 μg from a representative lot detected Jagged1 in one million Jurkat cells.

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Jagged1 in HT-29 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Jagged1 in HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound Jagged1 with a KD of 1.1 x 10-6 in an affinity binding assay.

Target description

Protein jagged-1 (UniProt: P78504; also known as Jagged1, hJ1, CD339) is encoded by the JAG1 (also known as JAGL1) gene (Gene ID: 182) in human. Jagged 1 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to generate the mature form that contains a large extracellular domain (aa 34-1067), a transmembrane domain (aa 1068-1093), and a cytoplasmic domain (aa 1094-1218). Jagged 1 is widely expressed in adult and fetal tissues. Its expression is observed in 32-52 days old embryos in the distal cardiac outflow tract and in pulmonary artery, pancreas, mesocardium, neural tube, and around the major bronchial branches. It serves as ligand for multiple Notch receptors and is involved in mediation of Notch signaling and cell-fate decisions during hematopoiesis. It is also reported to enhance FGF-induced angiogenesis. Its binding and activating ability are tightly regulated by Notch receptor glycosylation. Jagged 1 also plays roles in various aspects of cancer biology, including tumor angiogenesis, neoplastic cell growth, metastasis, epithelial-mesenchymal transition, and resistance to therapy. Its expression in other cells of tumor microenvironment has also been described. Jagged 1 expression can also be induced by other signaling pathways that are important in tumor growth and metastasis, including TGF-β, WNT/β-catenin, IL-6, and NF-kB. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
