General description

The immunocytochemical detection of bromodeoxyuridine (BrdU) incorporated into DNA is a powerful tool to study the cytokinetics of normal and neoplastic cells. In vitro or in vivo labeling of tumor cells with the thymidine analogue BrdU and the subsequent detection of incorporated BrdU with specific anti-BrdU monoclonal antibodies is an accurate and comprehensive method to quantitate the degree of DNA-synthesis. BrdU is incorporated into the newly synthezised DNA of the S-phase cells and can thus provide an estimate for the fraction of cells in S-phase. Also dynamic proliferative information (such as the S-phase transit rate and the potential doubling time) can be obtained, by means of bivariate BrdU/DNA flow cytometric analysis.

Specificity

Reacts specifically with bromodeoxyuridine. It also recognizes BrdU when incorporated into nuclear DNA.

Immunogen

A fusion of mouse myeloma cells with spleen cells from a mouse intraperitoneally immunized with BrdU conjugated to bovine serum albumin.

Application

Anti-BrdU Antibody, clone IIB5 is a Mouse Monoclonal Antibody for detection of Bromodeoxyuridine also known as BrdU & has been validated in FC, ICC, IHC(P).
Immunohistochemistry: 1:100-1:200 on frozen and paraffin embedded tissues with acid/proteinase treatments. See:

http://www.protocol-online.org/prot/Immunology/Immunohistochemistry. For examples of various protocols.

Flow Cytometry: 1:100-1:200; acid methanol treatment required.

Optimal working dilutions must be determined by the end user.

In order for the antibody to work a pretreatment to expose DNA will be needed (1 mg/mL of pepsin in 0.01 M HCL 30 min at r/t)

Mitophagy maintains tissue homeostasis by self-eliminating defective mitochondria through autophagy. How mitophagy regulates stem cell activity during hair regeneration remains unclear. Here, we found that mitophagy promotes the proliferation of hair germ (HG) cells by regulating glutathione (GSH) metabolism. First
