Gene Information

General description

Zyxin, is a cytosolic, focal adhesion protein belonging to the Ajuba family and containing 3 LIM zinc-binding domains. It associates with the actin cytoskeleton near the adhesion plaques and, during mitosis, with microtubules. Zyxin localizes to stress fibers, cell-cell junctions and colocalizes with vinculin, paxillin and VASP at focal adhesions but not at the tips of lamellipodia. Zyxin is involved in the regulation of actin dynamics and is a likely component of the signal transduction pathway mediating adhesion-stimulated changes in gene expression. Zyxin displays an actin nucleating activity that is dependent on its amino terminal proline-rich motifs and independent of Arp2/3 complex. Zyxin appears to function as an adaptor or docking protein to organize protein complexes important for regulated actin cytoskeleton dynamics.

Immunogen

KLH-conjugated linear peptide corresponding to human Zyxin.

Application

Immunocytochemistry Analysis: A 1:500 dilution from a previous lot detected Zyxin in NIH/3T3, A431, and HeLa cells.

This Anti-Zyxin Antibody, clone 14E11.1 is validated for use in WB, IC for the detection of Zyxin.

Quality

Evaluated by Western Blot in HUVEC cell lysate.

Western Blot Analysis: 0.5 µg/mL of this antibody detected Zyxin in 10 µg of HUVEC cell lysate.

Target description

~84 kDa observed. A non-specific band may be observed at ~36 kDa
The calculated molecular weight is 61 kDa Zyxin is a phosphoprotein with an apparent molecular weight of 82–84 kDa (van der Gaag, E., et al. (2002). Journal of Investigative Dermatology. 118:246–254.). As a substrate of caspases, Zyxin can be cleaved as 30, 37, 48 and 70 kDa (C-B Chan, C-B., et al. (2007). Cell Death and Differentiation. 14:1688–1699.)
