General description

Vacuolar protein sorting-associated protein 28 homolog (UniProt: Q9UK41; also known as H-Vps28, ESCRT-I complex subunit VPS28) is encoded by the VPS28 gene (Gene ID: 51160) in human. VPS28 is a component of the Endosomal Sorting Complex Required for Transport (ESCRT)-I complex that regulates the vesicular trafficking process. This complex forms the machinery driving protein sorting from endosomes to lysosomes. It is located in the cell membrane and late endosome membrane. Two isoforms of VPS28 have been described that are generated by alternative splicing. ESCRT complexes play a vital role in receptor down-regulation, lysosome biogenesis, and budding of HIV. In humans, ESCRT-I complex consists of TSG101, VPS28, and one of four potential human VPS37 homologues. The main role of ESCRT-I is to recognize ubiquitinated cargo via the UEV domain of the VPS23/TSG101 subunit. The N-terminal domain of VPS28 contains a conformationally dynamic hydrophobic binding site on its surface. The C-terminal domain of VPS28 is reported to bind the ESCRT-II complex. Western Blotting Analysis: A 1:1,000 dilution of this antibody detected recombinant GST-VPS28 protein.
