General description

Abelson interactor 1 (Abi1) protein, a 65 kD-substrate of the eponymous Abelson non-receptor tyrosine kinase (c-Abl) is expressed mainly in the central nervous system. This gene is located on human chromosome 10p12.
Abi 1 is an adaptor protein that interacts with Wave2 to regulate actin polymerization.

Immunogen

synthetic peptide corresponding to amino acids 163-180 of human ABI1, conjugated to KLH. The corresponding sequence is identical in several species including mouse, rat, and dog.

Application

Anti-ABI1 antibody produced in rabbit has been used in western blot analysis and immunofluorescence.
Rabbit anti- ABI1 antibody can be used for immunoblotting assays at a concentration of 0.5-1.0μg/ml using extracts from A431 and rat brain cells.

Biochem/physiol Actions

Abelson interactor 1 (Abi1) is essential for the formation and activation of WAVE2 signaling complex. ABI1 and WAVE are essential for Rac-dependent membrane protrusion and macropinocytosis, whereas ABI1 and N-WASP regulate actin-based vesicular transport, EGF receptor endocytosis, and EGFR and transferrin receptor (TfR) cell surface distribution. ABI1 has been shown to act as dual regulator of WAVE and neural Wiskott-Aldrich syndrome protein (N-WASP) specific activities. ABI1 is an essential component of this signaling complex. It positively regulates WAVE activity and connects WAVE to Rac through assembly of the WAVE-ABI1-Nap1-PIR121 complex. Abi1 modulates actin reorganization and lamellipodia formation.
