Mouse 129 teratocarcinoma AT805 derived
Cell Line Description
The chondrogenic cell line ATDC5 has been derived from teratocarcinoma AT805. Cells showed a sequential transition of phenotype in vitro. They encompass stages from mesenchymal condensation to calcification. Bone morphogenetic protein-2 (BMP-2) stimulated the sequential progression of early- and late-phase differentiation. Activation of parathyroid hormone (PTH)/PTH-related peptide (PTHrP) receptor resulted in inhibition of this differentiation capacity.
Application
ATDC5 has been used to study cartilage differentiation.
Culture Medium
DMEM: Ham′s F12 (1:1) + 2mM Glutamine + 5% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) i.e. seeding at 1-3 x 10,000 cells/cm² using 0.25% trypsin or trypsin/EDTA;CO2; 37°C subculture every 2-3 days.
