Gene Information

human ... ABCB5(340273)

General description

ABCB5 (ATP binding cassette subfamily B member 5) gene is located in human chromosome 7p21.1. The encoded protein belongs to the subfamily B multidrug resistance and ATP-binding cassette superfamily of active transporters. The encoded protein have two identical halves, each contains a hydrophobic region with six transmembrane segments and a nucleotide-binding region. Expression of this gene is observed in prostate and testis and in human melanoma tumor-initiating cells.
ABCB5 belongs to the ATP-binding cassette (ABC) transporter superfamily of integral membrane proteins. These proteins participate in ATP-dependent transmembrane transport of structurally diverse molecules ranging from small ions, sugars, and peptides to more complex organic molecules.

Immunogen

ABCB5 (NP_848654, 3-35)
This antibody is generated from rabbits immunized with a KLH conjugated synthetic peptide selected from the N-terminal region of human ABCB5.

Biochem/physiol Actions

ABCB5 (ATP binding cassette subfamily B member 5) is a cell surface marker for melanoma-initiating cells, on cellular metabolism. ABCB5 significantly regulates glycolysis, glutaminolysis and phospholipid metabolism. It induces stem cell differentiation, stem cell immortality and anti-apoptotic activity. The gene is upregulated in many cancer subtypes, including breast cancer. Recent studies reveal that ABCB5 is a novel marker for limbal stem cell and is needed for corneal development and repair. ABCB5 shows resistance against taxanes and anthracyclines, the anti-cancer drugs.

Physical form

Purified polyclonal antibody supplied in PBS with 0.09% (W/V) sodium azide.

Previously, we have demonstrated that human ABCB5 is a full-sized ATP-binding cassette transporter that shares strong homology with ABCB1/P-glycoprotein. ABCB5-transfected cells showed resistance to taxanes and anthracyclines. Herein, we further screened ABCB5 substrates, and explored the mechanism of resistance. Sensitivity
