General description

NMDA receptors are a class of ionotropic glutamate receptors. NMDA receptor channel has been shown to be involved in long term potentiation, an activity dependent increase in the efficiency of synaptic transmission thought to underlie certain kinds of memory and learning. NMDA receptor channels are heteromers composed of the key receptor subunit NMDAR1 (GRIN1) and 1 or more of the 4 NMDAR2 subunits: NMDAR2A (GRIN2A), NMDAR2B (GRIN2B or NR2B), NMDAR2C (GRIN2C), and NMDAR2D (GRIN2D). GRIN2B may be a candidate gene for the neurodegenerative disorder dentato-rubro-pallidoluysian atrophy (DRPLA). Properties of NMDAR include modulation by glycine, inhibition by Zn2+, voltage dependent Mg2+ blockade and high Ca2+ permeability. The involvement of NMDAR in the CNS has become a focus area for neurodegenerative diseases such as Alzheimer′s disease, epilepsy and ischemic neuronal cell death.

Specificity

NR2B; the immungen sequence is identical in mouse, rat, human, bovine, dog and chimp NR2B. Human NR2B is sometimes referred to as hNR3 in the literature.

Immunogen

a.a. 1437-1456 of mature mouse NR2B or 1463-1482 of NR2B precursor; signal is a.a. residues 1-26 [KFNGSSNG HVYEKLSSIESDV]. This sequence is identical to a.a. 1437-1456 of mature rat NR2B and a.a. 1465-1484 of human NR3, containing a N-terminal lysine.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Aliquot to avoid freezing and thawing. For maximum recovery of the product, centrifuge the original vial after thawing and prior to removing the cap.
Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
