Specificity

Clone C4 is a ZooMAb mouse recombinant monoclonal antibody that specifically detects Actin in most vertebrate species. It targets a conserved epitope within 20 amino acids from the N-terminal region.

Purified actin from chicken gizzard.

Application

Anti-Actin, clone C4 ZooMAb, Cat. No. ZMS1004, is a recombinant Mouse monoclonal antibody that targets Actin and is tested for use in Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Actin in HEK293, HeLa, and NIH3T3 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Actin in rat testis tissue sections.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected Actin in rat testis tissue sections.

Target description

Actin (UniProt: P68139) is encoded by the ACTA1 (also known as ACTA) gene (Gene ID: 421534) in chicken. Actins are ubiquitous eukaryotic proteins that serve as multi-functional, basic building blocks of cytoskeletal microfilaments. They play critical roles in a wide range of cellular processes, including cell division, cell migration, chromatin remodeling, transcriptional regulation, and vesicle trafficking. These functions are attributed to their ability to form filaments, which can quickly assemble and disassemble depending upon the needs of the cell. At least six different actin types have been reported in mammals. Although actins show about 90% overall sequence homology, isoforms do show spatial, temporal, and tissue-specific expression patterns and only 50-60% homology is found in their 18 N-terminal residues. Beta- and gamma-actins, also known as cytoplasmic actins, are highly conserved in higher animals and are predominantly expressed in non-muscle cells where they control cell structure and motility. They are nearly identical proteins and differ only in four amino acids at the N-terminal region. The other four actin isoforms are typically found in specific adult muscle tissue types. alpha-cardiac and alpha-skeletal actins are expressed in striated cardiac and skeletal muscles, respectively. Alpha and gamma actins are primarily found in vascular smooth muscle and enteric smooth muscles, respectively. Actin cytoskeleton is also involved in the morphogenesis of dendritic spines and is reported to be essential for spine structural stabilization that maintains long-term memory. Clone C4 is prepared as an antibody to chicken gizzard muscles actin, however, it reacts with actins from all vertebrates. It recognizes all six known vertebrate actins and binds to a highly conserved region of actin. This antibody reacts with both globular (G) and filamentous (F) forms of actin and does not interfere with actin polymerization to form filaments. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
