General description

The neurocalcin-δ (NCALD) gene is mapped to human chromosome 8q22.3. The encoded protein belongs to a large family of neuronal calcium (Ca2+) sensor (NCS) proteins and is a member of visinin-like protein (VILIP) subfamily. Neurocalcin-δ is also one of the EF-hand-containing Ca2+-binding proteins. It is predominantly expressed in the Purkinje cells of the cerebellum and is also present in the olfactory epithelium.

Specificity

Anti-Neurocalcin-d specifically recognizes human and mouse neurocalcin-d.

Immunogen

synthetic peptide corresponding to a sequence located near the N-terminus of human neurocalcin-δ, conjugated to KLH. The corresponding sequence is identical in mouse and rat neurocalcin-δ.

Application

Anti-Neurocalcin-δ antibody produced in rabbit is suitable for immunoblotting.

Biochem/physiol Actions

Neurocalcin-δ (VILIP-3) is a neuronal calcium (Ca2+) sensor. It binds cooperatively to three Ca2+ ions. Neurocalcin-δ influences cyclic guanosine monophosphate (cGMP) signaling in the retina, by acting as a regulator of the retinal guanylate cyclase (retGC1)/ photoreceptor membrane guanylyl cyclase 1 (ROSGC1). Neurocalcin-δ interacts with retGC1 in a calcium (Ca2+) dependent manner. It colocalizes with retGC1 in neurons in the inner plexiform layer of the retina, in amacrine and ganglion cells where it may influence synaptic signaling processes. Neurocalcin-δ has been shown to interact with actin, clathrin, and α- and β-tubulins, indicating that it may be involved with clathrin-coated vesicle traffic. It interacts and activates the olfactory guanylate cyclase ONE-GC, suggesting that it may function in regulating the odorant signaling pathway.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 1% BSA as stabilizer and 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
