General description

GRK6 (G protein-coupled receptor kinase 6) belongs to GRK family, and contains a central catalytic domain, which has N- and C-terminal on either sides. N-terminal is responsible for recognition of the receptor, and C-terminal is essential for plasma membrane binding of GRK6. This protein is composed of 576 amino acids, and has a molecular weight of 66kDa. This protein has a wide range of tissue expression.

Application

Biochem/physiol Actions

GRK6 (G protein-coupled receptor kinase 6) is involved in cell adhesion and locomotion, by phosphorylating and regulating chemokine receptor. It manipulates the organization of cytoskeleton and cell adhesion by controlling GRK-interacting protein (GIT1). It influences cAMP, epidermal growth factor receptor (EGFR) and calmodulin, thus, controlling the invasiveness and metastasis of tumor cells. It is up-regulated in hepatocellular carcinoma (HCC), and this is related to metastasis and prognosis of HCC. It has a major role in the phosphorylation of free fatty acid receptor 4 (FFA4), in a docosahexaenoic acid (DHA)-dependent manner. It regulates multiple leukocytes functions such as, activation of downstream effector signaling, receptor transport and localization at the cell membrane. This is achieved by controlling the involved CXCR2 protein, by forming a complex with AGS (activator of G-protein signaling)3-Gαi2. It is suppressed in medulloblastoma (MB), and this facilitates the migration of MB cells.
