Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TCF21 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TCF21 in A549 cell lysate.
Flow Cytometry Analysis: 0.1 µg from a representative lot detected TCF21 in one million A549 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TCF21 in Human kidney tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound TCF21 peptide with a KD of 3.1 x 10-7 in an affinity binding assay.

Transcription factor 21 (UniProt: O43680; also known as TCF-21, Capsulin, Class A basic helix-loop-helix protein 23, bHLHa23, Epicardin, Podocyte-expressed 1, Pod-1) is encoded by the TCF21 (also known as BHLHA23, POD1) gene (Gene ID: 6943) in human. TCF21 is a transcription factor of the basic helix-loop-helix (bHLH) family that is involved in epithelial-mesenchymal interactions in kidney and lung morphogenesis that include epithelial differentiation and branching morphogenesis. It bHLH domain is localized in amino acids 79-131. Its efficient DNA binding requires dimerization with another bHLH protein. It forms a heterodimer with TCF3 and binds the E box (5′-CANNTG-3′). TCF21 is specific for mesoderm and is expressed in the embryo mesenchymal derived tissues, such as the epicardium. It is reported to play a vital role in regulating cell differentiation and cell fate specificity through epithelial-mesenchymal transformation during cardiac development. TCF21 is shown to disrupt the central SRF/MYOCD interaction via multiple mechanisms, suggesting that it may also serve a master regulatory function in promoting smooth muscle cells de-differentiation. 外觀
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
