p16INK4A (cyclin-dependent kinase inhibitor 2A isoform) and p14ARF (cyclin-dependent kinase inhibitor 2A) are encoded by the CDKN2A (cyclin-dependent kinase inhibitor 2A, multiple tumor suppressor) gene. p16INK4A is a low molecular weight protein. This gene is mapped to human chromosome 9p21.

p16INK4A (cyclin-dependent kinase inhibitor 2A isoform) serves as a tumor suppressor and it negatively modulates cell growth by arresting cells in the G1 phase. Mutations in the CDKN2A (cyclin-dependent kinase inhibitor 2A, multiple tumor suppressor) gene is the main reason of inherited susceptibility to melanoma.
