應用
ACCUSPIN™无菌离心管(50 mL)搭配Histopaque™ -1077密度梯度分离液用于:
- 分离淋巴细胞等单个核细胞。
- 分离脐血–来源内皮祖细胞。
- 分离骨髓样品的单个核细胞层。
- 纯化人骨髓抽吸物,以研究BET抑制剂对急性髓系白血病的治疗价值。
原則
Histopaque-1077添加到ACCUSPIN管后,通过短暂离心将Histopaque-1077置于筛板下方。血液样品可以添加到管顶部腔体,而不会有与筛板下层腔体中Histopaque-1077混合的风险。在随后的离心中,全血下降通过筛板与Histopaque-1077接触。密度较大的组分取代Histopaque-1077的体积,使筛板上方清晰分离血液成分。该红细胞聚集,粒细胞变得轻微高渗,增加了其沉降速率,因此会沉淀到ACCUSPIN管底部淋巴细胞和其他单核的细胞,如单核细胞,保留在血浆/Histopaque-1077界面。这种单核细胞的密集带可以通过倾倒掉上层腔体中组分或使用移液管收集。红细胞污染可以被避免,因为腔体之间存在屏障。
