General description

Formaldehyde is widely employed in industry for a wide range of applications. It acts as a disinfectant and biocide, and is utilized as a tissue fixative and embalming agent.[1] Formaldehyde is a key metabolite produced during normal metabolism and contributes to methylation reactions and protein and nucleic acid synthesis. Increased production of formaldehyde has been observed in many cancer cell lines and increased formaldehyde concentration in urine has been associated with bladder and prostrate cancer.[2]

Principle

The Formaldehyde Assay kit provides a simple and direct procedure for measuring formaldehyde levels in urine and other biological samples. In this assay, formaldehyde is derivatized with acetoacetanilide in the presence of ammonia resulting in a fluorescent (λex = 370/λem = 470 nm) product, proportional to the formaldehyde present.

  1. Is the assay compatible with MES buffer? Is EDTA affecting the reaction?

    1. The principle of this kit is the reaction of formaldehyde with acetoacetanilide in the presence of ammonia. Neither a MESNA (4-Morpholineethanesulfonic acid, sodium) buffer nor EDTA in the test sample would be expected to interfere with this reaction. Having said that, it would be appropriate to run a blank containing MESNA and EDTA, and standards spiked with MESNA and EDTA, to confirm they they are not interfering with the test result...that is, making it either artificially high or low.

  2. What is the limit of detection that the MAK131 Formaldehyde Assay is capable of?

    1. The kit is capable of detecting formaldehyde down to 1.5 micromolar or 45 ppb. The upper end of detection for undiluted samples is 100 micromolar, which is the highest concentration of the standard curve.
