Gene Information

human ... CREB3L2(64764)

General description

CREB3L2 (cAMP responsive element binding protein 3-like 2) is also called BBF2 human homolog on chromosome 7 (BBF2H7), and along with old astrocyte specifically induced substance (OASIS) is a member of the cAMP response element-binding protein (CREB)/ATF (Activating transcription factor) family. It is a transmembrane protein, and acts as a bZIP (basic leucine zipper) transcription factor. This gene has 12 exons, and covers >120kbp. It has a transactivation region in its N-terminal, a leucine zipper, a DNA-binding domain, a transmembrane α-helical hydrophobic region, and a luminal domain. It is an endoplasmic reticulum (ER) protein, but an alternatively spliced variant is found in both ER and nucleus.

Application

Anti-CREB3L2 antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . Each antibody is tested by immunohistochemistry against hundreds of normal and disease tissues. These images can be viewed on the Human Protein Atlas (HPA) site by clicking on the Image Gallery link. The antibodies are also tested using immunofluorescence and western blotting. To view these protocols and other useful information about Prestige Antibodies and the HPA, visit sigma.com/prestige.

Biochem/physiol Actions

CREB3L2 (cAMP responsive element binding protein 3-like 2) confers protection during endoplasmic reticulum (ER) stress, and in response to ER stress is cleaved in its transmembrane region. It regulates the transcription of Sec23A, when it acts like a bZIP transcription factor. This is essential for collagen synthesis in dermal fibroblasts, where it ensures the proper trafficking of procollagen from ER-to-Golgi. It is a fusion partner of FUS gene in low-grade fibromyxoid sarcoma (LGFMS), and this fusion forms a supernumerary ring chromosome, which is related to tumor progression. It is a substrate for F-box and WD repeat domain-containing-7 (Fbxw7) protein, and is degraded by the same. This process, in turn, regulates osteogenesis and chondrogenesis. It also acts as a fusion partner of PPAR (Peroxisome proliferator-activated receptor) γ in thyroid carcinoma, and this fusion inactivates a thyroid signaling pathway.

Features and Benefits

Prestige Antibodies® are highly characterized and extensively validated antibodies with the added benefit of all available characterization data for each target being accessible via the Human Protein Atlas portal linked just below the product name at the top of this page. The uniqueness and low cross-reactivity of the Prestige Antibodies® to other proteins are due to a thorough selection of antigen regions, affinity purification, and stringent selection. Prestige antigen controls are available for every corresponding Prestige Antibody and can be found in the linkage section.

Every Prestige Antibody is tested in the following ways:
  • IHC tissue array of 44 normal human tissues and 20 of the most common cancer type tissues.
  • Protein array of 364 human recombinant protein fragments.
