Application

Alcian Blue 8GX has been used for the staining of sulfated proteoglycans in murine MPC (mesenchymal progenitor cell) nodule.[1] It has been used for the staining of sulfated proteoglycans in bovine articular chondrocytes.[2]

Biochem/physiol Actions

Alcian Blue 8GX is a cationic dye. It forms insoluble complexes with acidic glycosaminoglycans, thereby helping in the quantitative determination of glycosaminoglycans.[3] It is also used for the staining of bone and cartilage.[4]

Suitability

Certified for use in staining in Mowry′s pH 2.5 method, in Scott′s pH 5.7 method, and in Kreyberg′s method for keratin and mucin.

  1. Can the A9186 Alcian blue 8GX be used for the same assay as the A3157 Alcian blue 8GX previously used for mice spinal cord histology (frozen sections)?

    1. A9186 is intended to demonstrate glycoproteins when performing electrophoresis, while A3157 is intended for use with tissue sections. It is then tested on paraffin sections of small intestine, trachea, and larynx using different methods. Since A9186 was not tested for use with tissue sections, it is possible that it might not stain as well as product A3157, which was performance tested on paraffin sections. The same principle applies to other Alcian Blue 8GX products that are not tested for specific applications. It is advised to try using A9186, as the staining may be acceptable. If the staining is not acceptable, it is recommended to consider ordering A3157.
