Biochem/physiol Actions
Selective casein kinase 1 inhibitor. Potent inhibitor of histamine release from human basophils when induced by phorbol ester or DiC8, but not when induced by anti-IgE, fMLP (formyl-Met-Leu-Phe) and calcium ionophore A23187. This implicates divergent pathways of basophil activation.

The release of histamine and other inflammatory mediators from human basophils is triggered by numerous stimuli, including chemical, physical and receptor-mediated activators. Several mechanisms of cell activation including protein kinase C activation have been proposed to operate in these cells.

When screening various isoquinolinesulfonamide compounds which we synthesized, CKI-7, N-(2-amino-ethyl)-5-chloroisoquinoline-8-sulfonamide, was found to have a potent inhibitory action against casein kinase I and a much weaker effect on casein kinase II and other protein kinases. Kinetic analysis indicated that CKI-7
