General description

IgM normally constitutes about 10% of serum immunoglobulins. IgM antibody is prominent in early immune responses to most antigens and predominates in certain antibody responses such as ′natural′ blood group antibodies. IgM (with IgD) is the major immunoglobulin expressed on the surface of B cells. The gene for the mu constant region contains four domains separated by short intervening sequences.

Class specific anti immunoglobulin antibodies are useful for: The characterization of malignant B cell proliferations. All but acute lymphocytic leukemias share either surface or intra cytoplasmic Ig with an isotypic restriction, which suggest the monoclonal nature of the cell population. Most of the chronic lymphocytic leukemias, non Hodgkin lymphomas and Burkitt′s lymphoma bear surface IgM, whereas plasmocytes from Waldenström′s disease bear intracytoplasmic IgM. The other isotypes are less frequently found. On the other hand multiple myelomas are usually of the IgG or IgA type.

Specificity

Based on immunoelectrophoresis, the antibody reacts with the heavy chain portion of IgM. No reactivity with IgG or non-immunoglobulin serum proteins was detected, but cross reactivity with IgM from other species may be detected.

Physical form

Freeze dried powder, made from liquid containing 0.01M phosphate buffered saline, 0.25M sodium chloride, 15 mg/mL bovine serum with no preservatives. Reconstitute in 2mL deionized water, centrifuge if not clear. Prepare a working dilution fresh for each day.
ImmunoAffinity Purified

Storage and Stability

Maintain lyophilized product at 2-8°C for up to 12 months. After reconstitution the product is stable for several weeks at 2-8°C as an undiluted liquid. For extended storage after reconstitution, add an equal volume of glycerol to make a final concentration of 50% glycerol followed by storage at -20°C in undiluted aliquots for up to 12 months. Please note the concentration of protein (and buffer salts) will decrease to one-half of the original after the addition of glycerol. Avoid repeated freeze/thaw cycles.
