Hypoxia probe 2 is a resorufin-based non-fluorescent hypoxia probe that undergoes cytochrome P450 reductase (PH51)-catalyzed reduction to yield the fluorescent reporter resorufin (ex 545 nm, em 582 nm) in conditions of 4% O2 and lower, allowing imaging hypoxia levels similar to those detected by pimonidazole without the use of antibodies. When employed together with Tokyo green-based hypoxia probe 4 (Tokyo green ex 480 nm, em 515 nm), the probe pair allows the simultaneous (two-color) imaging of different hypoxia levels in living cells.
