General description

Transducin-like enhancer of split 3 (TLE3) belongs to the groucho family and comprises Q and WD40 domains. The TLE3 gene is mapped to human chromosome locus 15q23. It is expressed in adipocytes and shows higher levels in white adipose tissue (WAT) than the brown adipose tissue (BAT). It is also expressed in the prostate. TLE3 exists in isoforms due to alternative splice events.

Application

Biochem/physiol Actions

Transducin-like enhancer of split 3 (TLE3) acts as a modulator of adipogenesis. It is a transcriptional integrator for the nuclear receptor, peroxisome proliferator-activated receptor γ (PPARγ). TLE3 also interacts with Notch and controls the differentiation of epithelial cells. It serves as a transcriptional repressor for adenomatous polyposis coli (APC) and β-catenin in the wingless (Wnt) pathway by its interaction with the chromatin complex. TLE3 mediates inhibition of mitochondrial gene expression and early B-cell factor (EBF2) in beige adipocytes and regulates thermogenic gene expression. It may serve as a therapeutic target for obesity and diabetes.
