Specificity

Aquaporin 2 (AQP-CD/WCH-CD).

SPECIES REACTIVITY: The antibody has not yet been tested for cross-reactivity to other species. The 15 aa C-terminal peptide has 100% homology with mouse and 93% homology with human AQP2.

Immunogen

A 15 aa C-terminal peptide from rat AQP2. This peptide is predicted to be cytoplasmic.

Western Blotting: 1-10 μg/mL for affinity pure antibody using Chemiluminescence technique. An antibody made to the AQP2 peptide detected a major band at ~29 kDa in rat kidney cortex, and 35-47 kDa in inner and outer medulla (postranslational or glycosylation) (Nielsen et al. 1993; Deen et al. 1995). A similar pattern is seen in the human kidney (Sasaki, et al. 1994).

Immunohistochemistry: 2-10 μg/mL in paraformaldehyde fixed sections of tissues. AQP2 has been localized in apical membranes (Fushimi et al. 1993; Nielsen et al. 1993; Deen et al. 1995).

ELISA: 0.5-1.0 μg/mL using 1 μg/mL control peptide/well.

Optimal working dilutions must be determined by end user.

Physical form

Affinity purified immunoglobulin.Liquid in 100 mM Tris, pH 7.5 with 0.2% BSA and 0.05% sodium azide.

Storage and Stability

Maintain at -20°C in undiluted aliquots for up to 6 months after date of receipt. Avoid repeated freeze/thaw cycles.
