General description

Ionomycin is a calcium ionophore commonly derived from produced by the bacterium Streptomyces conglobatus, which means that it helps to transport calcium ions across cell membranes. It is a powerful tool for studying calcium signaling and its role in various biological processes, including cell activation, secretion, proliferation, and apoptosis.

Ionomycin is commonly used in cell biology experiments to induce a rapid and controlled increase in intracellular calcium levels. This allows to study the downstream effects of calcium signaling pathways, such as the activation of enzymes, the release of hormones, and the changes in gene expression.Ionomycin is also used to investigate the role of calcium in specific cellular processes, such as cell death, immune cell activation, and muscle contraction. For example, ionomycin can be used to induce apoptosis in cancer cells, study the activation of T cells, and investigate the role of calcium in muscle contraction.

Application

Ionomycin finds application in:
  • Study of calcium signaling and its role in various biological processes
  • Investigation of intracellular calcium levels, calcium-dependent enzymes, and cellular responses triggered by calcium influx
  • Induction of a rapid and controlled increase in intracellular calcium levels in cell biology experiments
  • Elucidation of specific cellular mechanisms and signaling pathways in combination with other substances, such as fluorescent calcium indicators or protein kinase activators
Ionomycin has been used:
  • to induce central demyelination[1]
  • inhibit adrenal bovine TREK-1 channels[2]
  • to regulate cell division of mature human B cells[3]

Biochem/physiol Actions

Ca2+ ionophore that is more effective than A23187 as a mobile ion carrier for Ca2+.
Ionomycin is an ionophoric antibiotic that binds Pb2+ and Ca2+, and serves as an effective mobile carrier of both cations.

Mode of Action: Facilitates the transfer of calcium ions (Ca2+) across biological membranes

Antimicrobial Spectrum: Active against Gram-Positive bacteria
Ionomycin is an ionophoric antibiotic which binds Pb2+ and Ca2+, and serves as an effective mobile carrier of both cations. Ionomycin is more effective than A23187 as a mobile ion carrier for Ca2+. It is non-fluorescent. It is used to study Ca2+ transport across biological membranes and induces apoptotic degeneration of embryonic cortical neurons.

Features and Benefits

  • Ready available solution reduce the need for preparation time
  • Efficient choice for studying calcium signaling and its role in various biological processes
  • Allows for rapid and controlled manipulation of intracellular calcium levels
  • Versatile and widely used in cell biology research

Questions

1–8 of 8 Questions
  1. Can this product be diluted (in DMSO) and stored -20C in single use aliquots? If so, what is the minimum concentration at which the product is still considered stable?

    1 answer
    1. This product is provided as 1 milliliter of a 1 millimolar (1 mM) solution of Ionomycin calcium salt. The solution may be thawed and aliquoted and re-frozen (e.g., 20 aliquots of 50 microliters each). But the recommendation would be to store the aliquots at 1 mM, and not to dilute further (in DMSO or anything else) before re-freezing.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.
For more information, please refer to the Product Dating Information document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/418/501/product-dating-information-06-25-mk.pdf

      Helpful?

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  4. What is the concentration of the Ionomycin calcium salt solution, Product No. I3909?

    1 answer
    1. It is a 1mM solution or ~ 0.75mg/mL in DMSO.

      Helpful?

  5. Is Product No. I3909 (Ionomycin calcium salt solution) hygroscopic?

    1 answer
    1. Yes, this product is hygroscopic.  It is packaged under an inert gas.  It is recommended to store I3909 under an inert gas and at -20°C protected from light.

  6. What is the Department of Transportation shipping information for this product?

    1 answer
    1. Transportation information can be found in Section 14 of the product's (M)SDS.To access the shipping information for this material, use the link on the product detail page for the product.

      Helpful?

  7. How does the storage temperature relate to shipping conditions?

    1 answer
    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

  8. Has Ionomycin calcium salt solution, Product No. I3909, been tested for use in Cell Culture applications?

    1 answer
    1. Sigma Aldrich has not tested this material for use in cell culture.  However, several references for use of ionomycin in cell culture applications can be found in the product information sheet (under Documents, above).
