  1. Is it possible to use Gomori Trichrome solutions as a replacement for Wheatley Trichrome solution when staining intestinal parasites?

    1. The Gomori Trichrome stain solution (Product HT10516) is not intended for staining intestinal parasites. It is specifically designed for use with paraffin sections of tissue to distinguish between muscle and collagen. The Gomori Trichrome procedure typically requires tissue fixation in Bouins Solution, but tissues are generally fixed in 10% Neutral Buffered Formalin and paraffin processed. On the other hand, the Wheatley Trichrome modification involves specimens that are typically fixed in PVA or Schaudinn's Fixative, the latter being a mercury-based fixative commonly listed in parasitology manuals. Due to the vastly different fixation procedures, there is no expectation that the Gomori Trichrome stain solution would effectively stain parasites.

  2. What is the expected slide yield from this kit?

    1. The number of slides that may be stained is highly variable. Typically, a volume of 40-50 ml of the stain solution is placed in a Coplin jar. A standard Coplin jar will hold 10-12 slides, depending on placement. Some larger staining jars include racks that hold up to 50 slides but require a significantly larger volume of stain solution. The stain solution may be used more than one time, however reuse is also impacted by the staining time required. If a few slides are stained each time and the stain is infrequently used, the number of repeat uses will be lower. Batch staining will result in the highest number of slides being stained per bottle. The stain solution should be changed whenever the user notices that the slide quality begins to diminish. Slides from each staining batch should be evaluated to ensure the staining results are acceptable. As staining quality begins to diminish, a fresh stain solution would be recommended. Do not return the used stain solution to the stock bottle.
