General description

Sigma′s 1 kb Ladder contains 11 fragments consisting of 500 bp repeats from 0.5 to 3 kb, 1 kb repeats from 3 to 6 kb, and 2 kb repeats from 6 to 10 kb. In nucleic acid gel electrophoresis, five μl of the marker should be diluted in gel loading buffer and then loaded in a single lane on an agarose or polyacrylamide gel. Suitable for use in Northern and Southern blotting. All DNA markers can be stained with ethidium bromide, SYBR Green, and Nancy-520.

Application

1 kb DNA Ladder has been used as a molecular marker to determine the molecular weight and size of double stranded DNA during gel electrophoresis.[1]

Other Notes

For optimal resolution, the recommended agarose gel concentration is 0.75%.
Sigma′s 1kb Ladder is provided in a solution of 10 mM Tris-HCl (pH 8.0), with 1.0 mM EDTA.

However, it is best to refer to the lot-specific Certificate of Origin, as the method of manufacture and origin of future lots may be subject to change.   2. how to use this product. in what ratio we have to add dye in this.

    1. This product has been tested successfully at 2 µL, 5µL, and 7 µL., however for general use, a starting volume of 5 µL is recommended. Dilute 5 µL volume of D0428 with 25 µL of gel loading buffer. Product G2526 is used for testing purposes. For best results, a 0.75% gel is recommended.
