General description

Amyloid β Protein is produced from amyloid-β precursor protein (APP). It consists of two C terminal variants, such as a long tailed Aβ 1-42 and a short tailed Aβ 1-40. APP is located on human chromosome 21q21.3.

Application

Amyloid β Protein Fragment 1-42 has been used:
  • for the preparation of Aβ 1-42 oligomer[1]
  • for western blot analysis
  • for the interference test of immunomagnetic reduction (IMR) plasma Aβ42 assay
  • to study the effects of resveratrol on Aβ 1-42-induced impairment of spatial learning, memory and synaptic plasticity
  • to investigate the effect of Aβ in epithelial cell culture

Biochem/physiol Actions

The predominant fragment of amyloid β-protein found in the brains of patients with Alzheimer′s disease and Down′s syndrome.
Amyloid β Protein Fragment 1-42 (Aβ 1-42) has antioxidant and neuroprotective properties. Accumulation of amyloid β Protein is associated with Alzheimer′s disease (AD) and Down Syndrome. Aβ 1-42 regulates cholesterol transport and may function as a transcription factor. It may possess anti-inflammatory and antimicrobial properties.

    1. This product can be reconstituted in ammonium hydroxide, pH >9, to a minimum of 1 mg/mL, or in DMSO at 0.4–4 mM.   2. After dissolving in DMSO, I want to dilute it with saline (finally 1ml/1mg) and use it for i.c.v. injection in mice. How many µl of 0.4-4 mM DMSO per 0.1 mg should I dissolve in?

    1. This product is soluble in DMSO at 0.4 - 4 mM, as stated in the product datasheet. This equates to 1.8 - 18 mg/mL in DMSO.   3. What is the difference between the AB peptide (1-42)(PP69) and AB protein fragment (1-42)(A9810)?

    1. These products are functionally the same, but are different brands. The PP69 is a Calbiochem product, while the A9810 is from Sigma-Aldrich. The testing specifications are slightly different, however both offer a minimum purity of 95%. The A9810 is lyophilized from a solution of water and TFA. The PP69 is lyophilized from a solution of 50 mM Tris-HCl. Please see the links below to review the product datasheet for each:

  4. It is instructed to dissolve the product in Ammonium Hydroxide solution, but what should be the molarity / Concentration of the ammonium hydroxide solution

    1. This product can be solubilized in ammonium hydroxide at a pH greater than 9. The concentration is not reported. It is recommended to prepare at a minimum 1 mg/mL concentration.

  5. How much ammonium hydroxide (pH >9) should be added to the 0,1 mg vial ? Is it possible to vortex the vial ?

    1. It is recommended to prepare a solution at a minimum of 1 mg/mL of ammonium hydroxide. Vertexing the vial is not necessary.

    1. As per the product information sheet, this product is soluble in ammonium hydroxide, pH >9, or in DMSO at 0.4–4 mM. Solutions can then be diluted into aqueous buffers such as10 mM sodium phosphate for use. Other reported solvents include hexafluoroisopropanol, acetonitrile, and acetic acid.
