General description

Tetramer molecular weight 465 kDa (subunits 116.3 kDa each)

Application

Aqueous solution may be used directly for derivatization without previous dialysis or gel permeation chromatography.

Biochem/physiol Actions

β-galactosidase cleaves lactose into its monosaccharide components, glucose and galactose. It also catalyses the transglycosylation of glucose into allolactose, the inducer of β-galactosidase, in a feedback loop.

Packaging

Bottomless glass bottle. Contents are inside inserted fused cone.

Other Notes

1 U corresponds to the amount of enzyme which hydrolyzes 1 μmol 2-nitrophenyl-β-D-galactopyranoside (Cat. No. 73660) per minute at pH 7.8 and 37 °C
Used in a highly sensitive enzyme-linked immunoassay of human IgG with 4-methylumbelliferyl-β-D-galactoside (Catalog No. 69590) [1]; Evaluation of β-D-galactosidase (E. coli) and horse radish peroxidase as labels by sandwich enzyme immunoassay technique [2]; Enzyme immunoassay of thyrptropin releasing hormone (TRH) [3]; Preparation of enzyme-antibody conjugates.

Questions

  1. I have already bought this one. My question is how will I reconstitute it? What diluent or buffer will I use?

    1 answer
    1. This product is lyophilized from a solution containing phosphate buffer and sucrose and may be reconstituted in water. For testing purposes, this product is dissolved at 5 mg/mL in a cold assay buffer. See below to review the composition of the 50 mM Tris assay buffer:
      1.0 ml 1 M Tris stock solution
0.4 ml of 0.5 M magnesium chloride stock solution
      0.01 ml Mercapto-ethanol
      Add to 15 ml H2O
      adjust pH 7.3 with HCl 1 M
      QS to 20 ml
