Specificity

Clone 4O15 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Vesicular glutamate transporter 1. It targets an epitope with in 19 amino acids from the C-terminal cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal region of rat Vesicular glutamate transporter 1 (VGlut1).

Application

Anti-VGluT1, clone 4O15 ZooMAb, Cat. No. ZRB2374, is a recombinant Rabbit Monoclonal Antibody that specifically targets Vesicular Glutamate Transporter 1 (aka Slc17a7 or VGlut1) and has been tested for use in Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Vesicular glutamate transporter 1 (UniProt: also known as VGluT1, Brain-specific Na(+)-dependent inorganic phosphate cotransporter, Solute carrier family 17 member 7) is encoded by the Slc17a7 (also known as Bnpi, Vglut1) gene (Gene ID: 116638) in rat. VGluT1 is a multi-pass synaptic vesicle membrane protein that mediates the uptake of glutamate into synaptic vesicles at presynaptic nerve terminals of excitatory neuronal cells. Glutamate transporters mediate glutamate uptake into synaptic vesicles and are driven by a proton electrochemical gradient generated by the vacuolar H+-ATPase. Two isoforms of VGluT1 have been reported that are produced by alternative splicing. VGluT1 is expressed in the cerebellum, cerebral cortex and hippocampus. Expression of isoform VGluT2 increases sharply during the first ten days of postnatal life, and also increases with increasing numbers of rhodopsin cells in the retina. It has been shown that both VGluT1 and VGluT2 are expressed prominently in the adult brain, however, during the first two weeks of postnatal development, VGluT2 expression predominates. VGluT1 expression becomes essential for survival at the time of the developmental switch from VGluT2 to VGLUT1. The VGluT1 knockout mice die at the age when a strong upregulation of VGluT1 through newly emerging VGluT1 synapses as well as a re+D4placement of the previously predominant VGLUT2 isoform in cerebellar, hippocampal, and cortical neurons occurs. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
