General description

There are 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. The five major classes of histones are H1/H5, H2A, H2B, H3, and H4. Histones H2A, H2B, H3 and H4 are known as the core histones, while histones H1 and H5 are known as the linker histones. Histones feature a main globular domain and a long N terminal tail which are involved with the structure of the nucleosomes of the ′beads on a string′ structure. They are the chief protein components of chromatin, acting as spools around which DNA winds, and play a role in gene regulation.

Specificity

Due to the highly conserved nature of histones, this antibody is predicted to have wide species reactivity.
The antibody binds specifically to an antigen determinant, which is present on all five histone proteins. The antibody recognizes the histones H1, H2A, H2B, H3 and H4 with comparable sensitivity.

Western Blot Analysis:
1:500 dilution of a previous lot was used.

Immunocytochemistry:
A 1-5 µg/mL concentration of a previous lot was used on Immunocytochemistry.

Immunohistochemistry:
A 1-5 μg/mL concentration of a previous lot was used on Immunohistochemistry.

Optimal working dilutions must be determined by end user.

Routinely evaluated by Western Blot on Jurkat lysates.

Storage and Stability

Stable for 6 months at 2-8ºC in undiluted aliquots from date of receipt.
The Iyophilized antibody is stable at 2–8°C. Store the reconstituted antibody in aliquots at -15 to -25°C for up to 12 months.Repeated freezing and thawing should be avoided.
