Application

Cell Counting Kit-8 (CCK-8) allows convenient assays using WST-8 (2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, monosodium salt), which produces a water-soluble formazan dye upon bioreduction in the presence of an electron carrier, 1-Methoxy PMS. CCK-8 solution is added directly to the cells, no pre-mixing of components is required. WST-8 is bioreduced by cellular dehydrogenases to an orange formazan product that is soluble in tissue culture medium. The amount of formazan produced is directly proportional to the number of living cells. Since the CCK-8 solution is very stable and it has little cytotoxicity, a longer incubation, such as 24 to 48 hours, is possible.
Cell Counting Kit-8 allows sensitive colorimetric assays for the determination of the number of viable cells in the proliferation and cytotoxicity assays. The detection sensitivity is higher than any other tetrazolium salts such as MTT, XTT or MTS.

  1. How, much volume to be used for 12 well plate. For e.g I am growing cells in my 12 well plate over a thin film. some paper mentions to use 10%, so if I add 100ul with 1ml media. will it work?

  3. 100 Test means 100 wells, so only one 96-well plate?

    1. The reagents provided in kit 96992 are sufficient for 100 tests. A 96 well microplate is not included with the kit. The customer must provide the 96 well plate. Although a solid 96 well plate can be used, a 96 well microplate with removable strips will allow for the most efficient use of the kit.

      •100 tests: 1 ml x 1 vial
      •500 tests: 5 ml x 1 vial
      •3000 tests: 5 ml x 6 vials

Based on the product datasheet, each well of a well-plate requires 10 microliters of solution, indicating that for 3000 tests, the kit should contain 30 ml of solution. Using these calculations, for the 100 test kit, each bottle would contain approximately 167 microliters of solution (5 ml divided by 30 = 167 microliters).

  6. Is this kit suitable for use with HEK293 or HEK293f cells?

    1. Yes, the 96992 kit can be utilized to stain both HEK293 and HEK293B cells. The Cell Count kit is not cell type-specific and is expected to function effectively for counting viable mammalian cells. In this kit, the WST-8 dye, which is a formazan stain is specific to viable cells and is not present in nonviable cells.
