Specificity

Clone 2D18 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CXCR2/IL-8RB. It targets an epitope within 25 amino acids from the N-terminal extracellular domain of the protein.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal extracellular domain of human CXCR2/IL-8RB.

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CXCR2/IL-8RB in HepG2 cell lysate.

Tested Applications

Affinity Binding Assay: The binding affinity of this antibody to CXCR2/IL-8RB peptide was at least five thousand-fold (5,000X) higher than that obtained with the non-specific control peptide.

Flow Cytometry Analysis: 1 µg from a representative lot detected CXCR2/IL-8RB in one million HepG2 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CXCR2/IL-8RB in HepG2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CXCR2/IL-8RB in human spleen tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

C-X-C chemokine receptor type 2 (UniProt: P25025; also known as CD182, CDw128b, GRO/MGSA receptor, High affinity interleukin-8 receptor B (IL-8R B), IL-8 receptor type 2) is encoded by the CXCR2C (also known as IL8RB) gene (Gene ID: 3579) in human. CXCR2/IL-8RB belongs to the G-protein coupled receptor 1 family. It is a membrane protein composed of seven transmembrane domains. It has a conserved DRY motif involved in G protein coupling. CXCR2 also contains N-linked glycosylation sites that can affect receptor trafficking and function. It primarily acts as a receptor for chemokines. Upon ligand binding, CXCR2/IL-8RB activates intracellular signaling pathways, including G protein-mediated signaling cascades and recruitment of β-arrestins. These signaling events regulate various cellular processes such as chemotaxis, neutrophil activation, angiogenesis, and immune cell recruitment. CXCR2/IL-8RB is expressed in a wide range of immune cells, including neutrophils, monocytes, T cells, and natural killer cells. It is also found in non-immune cells, such as endothelial cells and epithelial cells. The expression level of CXCR2 is predominantly induced during inflammation and infection. CXCR2 is primarily localized on the cell surface, where it functions as a membrane-bound receptor. It can also undergo internalization upon ligand binding, leading to receptor desensitization and subsequent downstream signaling events. Due to its role in immune regulation and inflammation, it has been associated with inflammatory conditions such as chronic obstructive pulmonary disease (COPD), asthma, rheumatoid arthritis, and inflammatory bowel disease. Additionally, CXCR2 has been implicated in cancer progression and metastasis by promoting angiogenesis, tumor growth, and metastatic cell migration. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
