Specificity

Clone 1O15 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human Proliferation marker protein Ki-67. It targets an epitope with in 14 amino acids from the C-terminal half.

Immunogen

KLH conjugated linear peptide corresponding to 14 amino acids from the C-terminal half of human Proliferation marker protein Ki-67.

Application

Anti-Ki67, clone 1O15 ZooMAb, Cat. No ZRB1007, is a recombinant Rabbit Monoclonal Antibody that specifically targets Proliferation marker protein Ki-67 and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Ki67 in HUVEC, HeLa, and A431 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:400 dilution from a representative lot detected Ki67 in human tonsil and human skin tissue sections.

Target description

Proliferation marker protein Ki-67 (UniProt: P46013; also known as Antigen identified by monoclonal antibody Ki-67, Antigen KI-67, Antigen Ki67) is encoded by the MKI67 gene (Gene ID: 4288) in human. Ki67 antigen is the prototypic cell cycle related nuclear protein, expressed by proliferating cells in all phases of the active cell cycle. Its expression occurs preferentially during late G1, S, G2 and M phases of the cell cycle, while in cells in G0 phase the antigen is undetectable. Ki67 is hyperphosphorylated in mitosis. Ki67 is required to maintain individual mitotic chromosomes and it disperses in the cytoplasm following nuclear envelope disassembly. It is shown to associate with the surface of the mitotic chromosome, the perichromosomal layer, and covers a substantial fraction of the chromosome surface. It prevents chromosomes from collapsing into a single chromatin mass by forming a steric and electrostatic charge barrier. Two isoforms of Ki67 have been described that are produced by alternative splicing. Ki67 antibodies are useful in establishing the cell growing fraction in neoplasms. In neoplastic tissues the prognostic value is comparable to the tritiated thymidine labelling index. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

120 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
