General description

INCENP or inner centromere protein antigens 135/155kDa is a constitutively binding centromere proteins and ′passenger,′ or transiently interacting, proteins. The overexpression of a truncation mutant of chicken INCENP and a chimeric CENPB:INCENP protein in mammalian cell culture disrupted both prometaphase chromosome alignment and completion of cytokinesis. INCENP forms a complex with the evolutionarily conserved family of Aurora B kinases which helps coordinate chromosome segregation, spindle behavior, and cytokinesis during mitosis.

Biochem/physiol Actions

As part of the chromosomal passenger complex (CPC), inner centromere protein (INCENP) has a role in assembly and stability of the complex. It also aids in the centromere localization of CPC. INCENP has a role in mitosis and associates with microtubules. The protein has been shown to be upregulated in colorectal cancer cell lines.

Physical form

Supplied in 50 mM sodium phosphate, pH 7.0, 300 mM NaCl, 150 mM imidazole, 0.1 mM PMSF, 0.2 mM DTT, 25% glycerol.

Preparation Note

After opening, aliquot into smaller quantities and store at -70 °C. Avoid repeating handling and multiple freeze/thaw cycles.

The inner centromere protein (INCENP), which has previously been described in chicken, frog and mouse, is required for correct chromosome segregation and cytokinesis. We have identified the human INCENP gene by library screening and reverse transcription-polymerase chain reaction (RT-PCR) and

Inner centromere protein (INCENP) is a chromosomal passenger protein with an essential role in mitosis. At the metaphase/anaphase transition, some INCENP transfers from the centromeres to the central spindle; the remainder then transfers to the equatorial cortex prior to cleavage
