General description

Persephin is a disulfide-linked homodimer neurotrophic factor structurally related to glial cell line-derived neurotrophic factor (GDNF), artemin, and neurturin. These proteins belong to the cysteine-knot family of growth factors that assume stable dimeric structures. Persephin signals through a multicomponent receptor system, composed of RET and one of four GFRa (a1-a4) receptors. The GFRa4 was first identified in chicken and was later shown to be the preferential binding subunit for Persephin. The gene coding for persephin is localized on human chromosome 19p13.3. Recombinant human persephin is a disulfide-linked homodimer, composed of two 10.3kDa polypeptide chains (192 total amino acid residues). Each chain contains seven conserved cysteine residues, one of which (Cys 63) is used for inter-chain disulfide bridging and the others are involved in intramolecular ring formation known as the cysteine knot configuration.

Biochem/physiol Actions

Persephin stimulates the survival of ventral midbrain dopaminergic neurons and motor neurons after sciatic nerve axotomy, and like GNDF, promotes ureteric bud branching. However, persephin does not support the survival of peripheral neurons.

Glial cell line-derived neurotrophic factor (GDNF) family ligands (GFL) are potent survival factors and regulators for central and peripheral neurons. GFLs bind to specific glycosyl phosphatidylinositol (GPI)-anchored co-receptors (GFRalpha1-alpha4), but signal through a common c-Ret receptor. Both GPI-anchored and soluble

Members of the GDNF protein family signal through receptors consisting of a GPI-linked GFRalpha subunit and the transmembrane tyrosine kinase Ret. Here we characterize the mouse Gfra4 and show that it undergoes developmentally regulated alternative splicing in several tissues. The
