General description

SCT262 is a novel fluorescent probe for the detection of ferrous ion (Fe2+) in mitochondria where Fe-S clusters and heme proteins are synthesized, and enables live cell fluorescent imaging of intracellular Fe2+. The SCT262 probe has no chelating ability. Add 53 uL DMSO to the 50 ug probe in the tube to create 1mM stock solution. Dilute with HBSS to prepare 5uM working solution.Cells may be incubated for 30 mins in the working solution, after which the supernatant solution can be replaced with the desired treatment solution.SPECTRAL PROPERTIESλex: 505 nmλem: 535 nmSuitable for use with FITC/GFP laser or filter

This probe is designed for the detection of mitochondrial ferrous iron in live cells by fluorescent microscopy or flow cytometry.

Target description

Iron is thought to be the most abundant transition metal element in many organisms, participating in diverse physiological activities. Recently, free iron in living cells has received attention due to its significance related to cellular damage or death. Free iron exists in its stable redox states, ferrous ion (Fe2+) and ferric ion (Fe3+). In living cells, understanding the behavior of Fe2+ is important because of the intracellular reductive environment, metal porters and water solubility of Fe2+. SCT262 is a novel fluorescent probe for the detection of ferrous ion (Fe2+) in mitochondria where Fe-S clusters and heme proteins are synthesized. This probe enables live cell fluorescent imaging of intracellular Fe2+. SCT262 has no no chelating ability. Mitochondrial FerroGreen and Fe2+ react irreversibly, which is different from the detection principle of calcium-iron probes such as Fluo-3.

Live cell probe for fluorescent detection of ferrous iron (Fe2+)in mitochondria by fluorescence microscopy or flow cytometry. SCT262 is optimized for the imaging of intracellular iron in live cell applications. IMPORTANT APPLICATION NOTE: Dissolve the lyophilized probe in in DMSO to make a 1 mM stock solution, and dilute with HBSS or serum-free medium to make a 5 uM immediately before staining. Use DMSO-based stock solution same day. Alternatively, probe dissolved in ethanol is stable at -20C for two weeks.
