General description

CCL2, Chemokine (C-C motif) ligand 2, or alternatively known as Monocyte chemotactic protein 1 (MCP-1), Monocyte chemotactic and activating factor (MCAF), Monocyte chemoattractant protein 1, Monocyte secretory protein JE, Small-inducible cytokine A2, HC11, and C-C motif chemokine 2 is encoded by the human gene named CCL2/MCP1/SCYA2. CCL2 is a chemokine produced by endothelial cells that attracts memory T cells, monocytes, dendritic cells, and basophils but not neutrophils or eosinophils to sites of inflammation produced by either tissue inquiry or infection. CCL2 also bolsters monocytes anti-tumor activity, and because it is chemotactic for monocytes, CCL2 has been implicated in inflammatory diseases where monocytic invasions have occurred such as in psoriasis, rheumatoid arthritis and atherosclerosis. CCL2 is a bifunctional molecule as well; the whole molecule is a basophil activator but removal of the N-terminal portion converts the molecule into an exclusive eosinophil chemoattractant. CCL2 can also be found expressed at the sites of tooth eruption and bone degradation and is expressed by both osteoclasts and osteoblasts. CCL2 is also expressed in neurons, astrocytes and microglia and its role in neuroinflammation is established. CCL2 expression in glial and neurons is increased in epilepsy, brain ischemia, Alzheimer’s disease, brain injury and experimental autoimmune encephalomyelitis. Finally, CCL2 plays a role in vascular complications associated with type II diabetes and in the production of amylin which contributes to insulin resistance in obesity, which is also associated with type II diabetes. EMD-Millipore’s Anti-CCL2 mouse monoclonal antibody has been tested in western blot against recombinant protein and cell lysates from A549, HeLa, Raw264.7, L1210, C6, and COS7 cell lines.

Application

Detect Monocyte Chemotactic Protein using this mouse monoclonal antibody, Anti-CCL2 Antibody, clone 2D8 validated for use in western blotting, Immunofluorescence, IHC & Flow Cytometry.
Immunofluorescence Analysis: A 1:200-1,000 dilution from a representative lot detected CCL2 in HeLa cells.

Immunohistochemistry Analysis: A 1:200-1,000 dilution from a representative lot detected CCL2 in human liver cancer tissue.

Flow Cytometry Analysis: A 1:200-400 dilution from a representative lot detected CCL2 in A549 cells.

Optimal working dilutions must be determined by end user.
