General description

53kDa excretory/secretory antigen (UniProt: Q27071; also known as 53 kDa excretory/secretory antigen, gp53) is encoded by the gp53 gene (Gene ID: 10908901) in Trichinella spiralis. Trichinells species are parasites that cause trichinellosis resulting from the consumption of undercooked or raw meat. Its life cycle is relatively short during which it undergoes four molts to reach adult state. The newborn larvae produced by females in the intestine become lodged in striated muscle fibers where they can stay for long periods of time. The gp53 glycoprotein excreted and secreted by larvae serve as a an immunodiagnostic candidate for detection of trichinellosis. It is produced and glycosylated in β-stichocytes. It has three glycosylation sites (Asn 146, 279, and 328) and contains six phosphorylation sites, and three consensus myristoylation sites. Clone US9 is reported to recognize gp53 in all encysting Trichinella species. It recognizes a linear epitope of eight residues (Met33-Ser40; MTFSVPIS) localized in the N-terminal region. The recognition of this epitope has been attributed to the availability of hydroxyl groups provided by methionine and serine on gp53. (Ref.: Orbegozo-Medina, RA et al., (2019). Specificity

Clone US9 is a mouse monoclonal antibody that specifically detects Trichinella gp53 protein. It targets an epitope within 8 amino acids from the N-terminal region.

Immunogen

Spleen cells from Trichinella spiralis-infected male NBF1 mice fused with P3-X63-Ag8.653 B lymphoblast cells.

Application

Quality Control Testing

Evaluated by Western Blotting with a chimeric peptide containing epitope MTFSVPIS.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected the epitope MTFSVPIS within a chimeric peptide of 50 amino acids from Trichinella gp53.

Tested Applications

Western Blotting Analysis: A representative lot detected Trichinella gp53 in Western Blotting applications (Romaris, F., et al. (2002). Mol Biochem Parasitol. 125(1-2):173-83).

ELISA Analysis: Serial dilutions from a representative lot detected Trichinella gp53 in T1 chimeric peptide with epitope sequence MTFSVPIS.

Immunohistochemistry Applications: A representative lot detected Trichinella gp53 in Immunohistochemistry applications (Romaris, F., et al. (2002). Mol Biochem Parasitol.125(1-2):173-83).

ELISA Analysis: A representative lot detected Trichinella gp53 in ELISA applications (Romaris, F., et al. (2002). Mol Biochem Parasitol.125(1-2):173-83).

Target description

6 kDa observed for T1 peptide; 46.64 kDa calculated for the complete gp53 protein. Uncharacterized bands may be observed in some lysate(s).

Purified mouse monoclonal antibody IgG1 in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.
