Gene Information

Specificity

Specific for Dopamine Receptor D4. Mouse Dopamine receptor D4 is a 387 aa, G-protein coupled and transmembrane receptor protein (Van Tol et al. 1992; Legerski & Peterson 1992; Livingstone et al. 1992; Seeman et al. 1994). The immunogen peptide shows no significant homology with other dopamine receptors (D1, D2, D3 or D5).

SPECIES REACTIVITIES: The immunogen peptide shows sequence homology of 90% with rat and human D4 receptors (Van Tol et al. 1992; Legerski & Peterson 1992; Livingstone et al. 1992; Seeman et al. 1994; O′Malley et al. 1992). Antibody cross-reactivity with D4R from various species is not known.

Immunogen

A 10 amino acid peptide sequence from the mouse D4 receptor within the 3rd extracellular domain.
Epitope: extracellular domain

Application

Anti-Dopamine D4 Receptor Antibody, extracellular domain is an antibody against Dopamine D4 Receptor for use in ELISA & WB.

Western blot: 1-10 μg/mL (Chemiluminescence technique). Approximately 100-400 μg of membrane protein/lane should be loaded to visualize the receptor band. Antibody detects ~55kDa band in HT222 cells and mouse hippocampal membranes. {http://www.jneurosci.org/cgi/reprint/21/16/6069.pdf, Ishige, K et.al.(2001) J. Neuroscience 21(16):6069-6076.

ELISA: 0.5-1.0 μg/mL (1 μg/mL immunogen peptide/well)

Not tested in immunohistochemical methods.

Optimal working dilutions must be determined by the end user.

Pharmacological studies suggest that dopamine release from lateral olivocochlear efferent neurons suppresses spontaneous and sound-evoked activity in cochlear nerve fibers and helps control noise-induced excitotoxicity; however, the literature on cochlear expression and localization of dopamine receptors is contradictory.
