Specificity

Clone 1G16 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects SDHA. It targets an epitope within 19 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal half of human Succinate dehydrogenase [ubiquinone] flavoprotein subunit (SDHA).

Application

Quality Control Testing

Evaluated by Western Blotting in RAW264.7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SDHA in RAW264.7 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected SDHA in MCF-7, C2C12, and HepG2 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SDHA in HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected SDHA in human heart tissue sections.

Target description

Succinate dehydrogenase [ubiquinone] flavoprotein subunit, mitochondrial (UniProt: P31040; also known as EC:1.3.5.1, Flavoprotein subunit of complex II, Fp, SDH-Fp) is encoded by the SDHA (also known as SDH2, SDHF) gene (Gene ID: 6389) in human. SDH-Fp is a member of the FAD-dependent oxidoreductase 2 family that is localized in the inner mitochondrial membrane. It is the flavoprotein subunit of succinate dehydrogenase that is involved in complex II of the mitochondrial electron transport chain and is responsible for transferring electrons from succinate to ubiquinone (coenzyme Q). Complex II is composed of four nuclear-encoded subunits: the flavoprotein (FP) SDHA, iron-sulfur protein (IP) SDHB, and a cytochrome b560 composed of SDHC and SDHD. SDH-Fp undergoes phosphorylation at tyrosine 215 that is shown to be important for efficient electron transfer in complex II and for the prevention of reactive oxygen species (ROS) generation. Three isoforms of SDH-Fp have been described that are produced by alternative splicing. Mutations in SDHA gene are known to cause mitochondrial complex II deficiency, nuclear type 1 (MC2DN1) that is characterized by psychomotor regression in infants, poor growth with lack of speech development, muscle weakness, exercise intolerance, and cardiomyopathy. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
