General description

Imidazole is used as a buffer in the pH range of 6.2-7.8 at 25°C. It is recommended as a component of a buffer for the assay of horseradish peroxidase and as a chelator for the binding of various divalent cations. Imidazole aids the elution of histidine-containing proteins from divalent cation resins (Sigma P6611, HisSelect.-HC Nickel affinity gel) and can also be used in reverse staining of SDS-PAGE gels for the detection of proteins.

Application

Imidazole buffer solution has been used as a component:
  • of calmodulin-binding buffer 1 (CBB1) for the tandem affinity purification (TAP) of TAP-tagged proteins together with co-purifying proteins
  • of lysis buffer, and SDS loading buffer for His-ubiquitin pull-down assays
  • of imidazole wash and elution buffer for protein purification
