Application

Hemin is used for the following applications:
  • To study bacterial strains and growth conditions[1]
  • Used in short-term colony assays (the culture medium consists of hemin along with other components)[2]
  • Cell transfections[3]
It may be used in β-hematin formation assay for analyzing the inhibitory activity of MMV (Medicines for Malaria Venture) Malaria Box compounds against the formation of β-hematin.[4]

Biochem/physiol Actions

Hemin, the oxidized version of heme, is an iron-containing prosthetic group for a diverse group of proteins. Free heme, which can be released from hemoglobin following hemolysis, is pro-inflammatory and contributes to iron-derived reactive oxygen species. Free hemin levels may be upregulated in various pathological conditions and can contribute to various inflammatory conditions including vascular disorders, renal failure, and immune-mediated disorders.

Hemin may be used to study the expression and represson of heme oxygenase 1 (HO-1) as well as the activity of HO-1. Hematin is the "X factor" required to grown Haemophilus influenzae.

  1. Can 51280 be used to induce the erythroid differentiation of the K562 cells? If yes, what would it need to be dissolved in to add be added to cell culture?

    1. This product is not tested for any specific application. However, the material has been used successfully by other researchers to induce erythroid differentiation and hemoglobinization of K562 cells. Please see the link below to review one such publication:

      For culture use, this compound is typically prepared in DMSO at a concentration of 1 mM. The material is also soluble in 1.4 M ammonium hydroxide at 24 mg/mL at ambient temperature or 50 mg/ml when heated to 95°C for 5 minutes. Additionally, stock solutions of 0.25% hemin in 50% triethanolamine have been reported. Published methods suggest rates of 40 - 60 µM hemin for a treatment period of 1-5 days. Filter-sterilized stock solutions may be stored refrigerated, protected from light.

If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life.     1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. It is also soluble in 1.4 M ammonium hydroxide at a concentration of 50 mg/ml, and this can be achieved by heating the solution at 95°C for 5 minutes, resulting in a dark brown solution. Additionally, a stock solution of 0.25% hemin in 50% triethanolamine can be prepared. CAS number 16009-13-5, which corresponds to product 51280, is soluble in 1.4 M NaOH at a concentration of 25 mg/mL, and it is practically insoluble in water.

  5. How can hemin be dissolved for cell culture experiments in macrophages? Is it necessary to couple it to serum/BSA?

    1. This product has not been tested for use in cell culture. However, a previously deleted product, H1652, has been tested. The H1652 was dissolved at 10 mg/mL in 1 M NaOH and added then added to the culture medium at 7 mg/L. This information would need to be validated for product 51280.
