General description

Inhibin alpha chain (UniProt: P04087; also known as IHNA, A-inhibin subunit) is encoded by the INHA gene (Gene ID: 397386) in porcine. Inhibins are dimeric glycoproteins that are composed of an -subunit and either a βA (inhibin A) or a βB-subunit (inhibin B). Inhibins and activins inhibit and activate, respectively, the secretion of follicle stimulating hormone (FSH) by the pituitary gland. They are also involved in regulating a number of other functions. Inhibins are glycoprotein hormones of the TGFβ superfamily and are composed of two subunits, an -subunit (20 kDa) and a β-subunit (13 kDa), linked by a disulfide bridge. Two forms of inhibin have been identified, Inhibin-A and -B. Their subunits are shown to be identical, while the β-subunit differs between both forms of inhibin and are denoted as βA and βB, respectively. Inhibin-A is secreted during the luteal phase, while Inhibin-B is predominantly secreted during the follicular phase. The inhibin -subunit contains an N-linked glycosylation site and this glycosylation is shown to be essential for its FSH suppressing activity. Porcine Inhibin chain is synthesized as a protein of 364 amino acids with a signal peptide (aa 1-17) and a propeptide (aa 18-60), which are subsequently cleaved off to generate Inhibin N-terminal region (aa 61-230) and the Inhibin chain (aa 231-364). Subsequent cleavage at aa 230-231 results in the production of Inhibin chain. Circulating level of Inhibins is a reliable marker for some types of ovarian cancer and is a predictor for the conditions of spermatogenesis. (Ref.: Makanji, Y., et al. (2014). Endocr. Rev. 35(5); 747-794; Bilezikjian, LM., et al. (1993). Endocrinology. 133(6); 2545-2553).

Specificity

This rabbit polyclonal antibody detects Inhibin chain. It targets an epitope within the first 26 amino acids from the mature Inhibin chain.

Immunogen

Linear peptide corresponding to the first 26 amino acids from the mature Inhibin chain of Porcine (aa 231-256).

Application

Quality Control Testing

Evaluated by Immunohistochemistry (Paraffin) in porcine testis tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution of this antibody detected Inhibin chain (INHA) in porcine testis tissue sections.

Tested Applications

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
Anti-Inhibin chain (INHA), Cat. No. ABS2207, is a rabbit polyclonal antibody that detects Inhibin a chain and is tested for use in Immunohistochemistry (Paraffin) and Western Blotting.

Physical form

Rabbit polyclonal antibody in serum with 0.05% sodium azide.

Storage and Stability

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
